Physical pellet quality and AMEN concentration are strongly related to each other in broiler feeding. A study was conducted to evaluate the relationship between dietary AMEN concentration and feed processing on pellet quality, nutrient digestibility, broiler performance, serum markers, and yield of commercial cuts. Six diets were formulated. The first diet had the recommended AMEN concentration, each further diet was calculated with 40 kcal/kg less, from 0 to −200 kcal/kg, resulting in six levels for each feed phase: starter (1−14 d), grower (15−28 d), and finisher (29−35 d). These diets were processed with and without expander conditioning prior to pelleting, using an average corn particle size of 1.6 mm, ground with a roller mill. A total of 1008 one-day-old male Ross 308 broiler chickens were placed in a 6 × 2 (6 energy levels and 2 conditionings) factorial trial with six boxes as replications, with three in each broiler performance trial period. Excreta were collected 2 days before the end of each feed phase for apparent total tract digestibility measurement. On day 36, four broilers from each replication (pen) were weighed and then euthanized for blood collection, following which the gastrointestinal organs were weighed, and the ileal and gizzard contents were collected. On day 37, all remaining broilers were slaughtered after fasting to measure commercial cuts and abdominal fat. The results show that the pellet durability index (PDI) was most affected by energy reducing and expander conditioning prior to pelleting, and it was better when diets had energy reduced by 40 to 200 kcal/kg (p > 0.001), as when expander conditioning was used. Digestibility of nutrients was slightly affected by treatments, as was the broiler performance; however, feed efficiency was improved in broiler-fed diets without AMEN reduction and when an expander was used, with p = 0.050 and p = 0.031, respectively. No effects were observed on the weight of gastrointestinal tract organs and serum markers, except for liver (p = 0.037) and α-amylase (p = 0.047). The lowest liver weight and lowest serum protein, cholesterol, triglyceride, gamma-glutamyl, and lipase concentrations were obtained when diets were formulated without energy reduction (Ross-0). There was no effect on commercial cuts relative to live weight at slaughter. The energy reduction was well reflected in the proportion of abdominal fat, which decreased when AMEN was reduced (p = 0.001). The present study shows it is possible to use diets with up to 200 kcal/kg reduction in AMEN without losses in performance, and the use of expander conditioning prior to pelleting promotes higher pellet quality and broiler feed efficiency.

During the processing of compound feed for broilers, several changes occur that affect the physical and probably the nutritional properties of pellets, influencing animal performance. The effects of mill type, particle size (PS) and expander conditioning prior to pelleting (E + P) were combined to generate pellets. A 2 × 3 × 2 factorial arrangement was designed with two mill types (a hammer mill (HM) or roller mill (RM)), three PSs (0.8, 1.2 or 1.6 mm) and two E + Ps (with or without expander processing prior to pelleting), with six replications of 12 unsexed Ross 308 broilers each. All the processing lines reduced the PS from mash to finished pellets via secondary grinding, by 2.35 times on average. However, RM grinding required less electric power (p < 0.001). The intended PS (0.8, 1.2 or 1.6 mm) did not affect this energy consumption. E + P and the PS interacted for the pellet durability index (PDI) (p = 0.006). The worst PDI in the pellets was observed when a PS of 1.6 mm without E + P was used. Only E + P positively affected starch (p < 0.001) and amino acids' ileal apparent digestibility (p < 0.01). Organic matter (OM) (p = 0.02) and fat (p < 0.001) digestibility, as well as AMEN (p = 0.005) content, were influenced by the PS (main effect), whereas E + P and mill type interacted with these values (p < 0.005). Lower OM digestibility and AMEN content were observed when RM without E + P was used (p = 0.001). The feed conversion ratio (FCR) was enhanced and feed intake (FI) was improved with E + P. The combination of the RM mill, a 1.6 mm mean PS, and E + P improved FCR (three-way interaction, p = 0.019)), showing that for a higher PS, E + P is necessary for animal performance. Carcass yield was, on average, 80.1%. No effects on commercial cuts (breast, legs and wings) were observed. In contrast, abdominal fat was affected by mill type * PS (p = 0.012) and E + P * PS (p = 0.048) in a two-way interaction. The highest abdominal fat indicated an imbalance in the amino acid (AA)-to-AMEN ratio. Coarse PS promoted heavier gizzards (p = 0.02) but E + P tended to reduce them (p = 0.057). The processing steps improved pellet quality and feed efficiency associated with RM, coarse PS and E + P, highlighting the positive effects of E + P on abdominal fat and AMEN content, which should be adjusted to AA or reduced at formulation. However, these results are for an experimental processing plant and may not necessarily apply to larger plants, so the use of these data and methods should be considered as guidelines for replication at production sites.

When it comes to the investigation of key ecosystems in the world, we often omit salt from the ecological recipe. In fact, despite occupying almost half of the volume of inland waters and providing crucial services to humanity and nature, inland saline ecosystems are often overlooked in discussions regarding the preservation of global aquatic resources of our planet. As a result, our knowledge of the biological and geochemical dynamics shaping these environments remains incomplete and we are hesitant in framing effective protective strategies against the increasing natural and anthropogenic threats faced by such habitats. Hypersaline lakes, water bodies where the concentration of salt exceeds 35 g/l, occur mainly in arid and semiarid areas resulting from hydrological imbalances triggering the accumulation of salts over time. Often considered the 'exotic siblings' within the family of inland waters, these ecosystems host some of the most extremophile communities worldwide and provide essential habitats for waterbirds and many other organisms in already water-stressed regions. These systems are often highlighted as natural laboratories, ideal for addressing central ecological questions due to their relatively low complexity and simple food web structures. However, recent studies on the biogeochemical mechanisms framing hypersaline communities have challenged this archetype, arguing that newly discovered highly diverse communities are characterised by specific trophic interactions shaped by high levels of specialisation. The main goal of this review is to explore our current understanding of the ecological dynamics of hypersaline ecosystems by addressing four main research questions: (i) why are hypersaline lakes unique from a biological and geochemical perspective; (ii) which biota inhabit these ecosystems and how have they adapted to the high salt conditions; (iii) how do we protect biodiversity from increasing natural and anthropogenic threats; and (iv) which scientific tools will help us preserve hypersaline ecosystems in the future? First, we focus on the ecological characterisation of hypersaline ecosystems, illustrate hydrogeochemical dynamics regulating such environments, and outline key ecoregions supporting hypersaline systems across the globe. Second, we depict the diversity and functional aspects of key taxa found in hypersaline lakes, from microorganisms to plants, invertebrates, waterbirds and upper trophic levels. Next, we describe ecosystem services and discuss possible conservation guidelines. Finally, we outline how cutting-edge technologies can provide new insights into the study of hypersaline ecology. Overall, this review sheds further light onto these understudied ecosystems, largely unrecognised as important sources of unique biological and functional diversity. We provide perspectives for key future research avenues, and advocate that the conservation of hypersaline lakes should not be taken with 'a grain of salt'.

Two meat-type broiler lines, line A and line B were fed experimental diets from 22-42 d with objectives to determine the effects of dietary metabolizable energy (ME) levels on feed intake (FI), performance, body composition, and processing yield as affected by environmental grow-out temperatures. Two thousand fifty male chicks from line A and 2,050 male chicks from line B were reared in 90-floor pens, 45 chicks per pen utilizing primary breeder nutrition and husbandry guidelines for starter (1-10 d) and grower (11-21 d) phases. Experimental finisher diets consisted of 5 increasing levels of apparent nitrogen corrected ME (2,800, 2,925, 3,050, 3,175, and 3,300 kcal/kg set at 19.5% crude protein and 1.0% dLys at each level) to represent 80, 90, 100, 110, and 120% ME of Evonik AminoChick energy level giving 2 × 5 factorial design and were fed from 22-42 d. All other amino acid levels in diets were formulated to a fixed ratio of dLys level. There were nine replicate pens for each diet and each line. The experiment was conducted twice-once in hot season (barn averages: 77.55 ˚F and 86.04% RH) and another in cool season (barn averages: 69.91 ˚F and 63.98% RH) of the year. Results showed that FI and feed conversion ratios (FCR) decreased (P < 0.05) linearly (R2 = 0.9) by 61.25 g and 0.073 units for every 10% increase in dietary ME for combined analysis of lines and seasons. The % fat mass of total body mass increased by 0.57%, whereas % protein mass decreased by 0.21% across ME levels (R2 > 0.9). However, there was no difference (P > 0.05) in % weights (of live weight) for wings, breast filet, tenders, or leg quarters across ME levels for both lines except % fat pad that increased (P < 0.05) by 0.20% for each 10% increment in dietary ME level. Line B had higher cumulative FI, BW gain, % lean, and protein mass of body mass than line A in hot season (P < 0.05). Feed intake was not different between lines in cool season (P > 0.05), whereas higher BW and improved FCR were observed for line A. Line A had higher % fat mass in both seasons. In summary, performance and yield results as affected by dietary ME levels were line specific and were affected by grow-out seasons. The optimal dietary ME level for the ME range studied (2,800-3,000 kcal/kg) at a constant recommended amino acid level lies in determining the best performance and profitability indices by taking into account the grow-out production inputs and processing yield outputs.

Knowledge about the nutritional value of methionine sources is highly important for their appropriate application in terms of animal and economic performance. Therefore, a broiler feeding trial was conducted to determine the relative bioavailability value (RBV) of DL-2-hydroxy-4-methylthio butanoic acid (HMTBA) compared to DL-methionine (DLM). DLM diluted to 65% purity (DLM65) served as the internal standard, with a known RBV of 65%. A total of 1920 d-old male broilers were used in the three-phase feeding trial comprising 16 treatments including a basal, Met+Cys-deficient diet and 5 graded DLM, HMTBA, or DLM65 levels. Growth performance and carcass quality data were subjected to multi-exponential regression analysis. Increasing levels of any Met source significantly improved all performance parameters compared to the negative control (p < 0.05). Across all performance parameters, the RBV of HMTBA was 63% and that of DLM65 was 58%. All RBV estimates of HMTBA and DLM65 were significantly lower than 88% (p < 0.05). Cumulative efficiency of DLM for Met+Cys deposition in body protein was higher than that of HMBTA at any dose, confirming the determined RBV. Using DLM65 as an internal marker allowed for validation of the methodology.

Reducing dietary CP for broiler chickens below a certain threshold results in decreased growth, even when the supply of essential amino acids and glycine equivalent (Glyequi) is adequate, probably because other nonessential amino acids (neAA) are growth-limiting. Nonprotein nitrogen (NPN) might be used for the synthesis of neAA. Therefore, the effects of specific neAA and ammonium chloride (NH4Cl) supplementation on the growth and N-excretion characteristics of broiler chickens were investigated. Nine male Ross 308 broiler chickens were kept in each of 81 metabolism units from day 7 to 21 and received 1 of 9 diets in 9 replicates in a one-factorial arrangement of treatments. Two diets with different neAA concentrations, except for Glyequi, were mixed resulting in CP levels of 180 (CP180) and 160 (CP160) g/kg. In six other diets, CP160 was supplemented with either l-Ala, l-Pro, l-Asp, a mix of l-Asp and l-Asn·H2O, l-Glu, or a mix of l-Glu and l-Gln to achieve concentrations of the respective neAA as formulated in CP180. In a further diet, NH4Cl was added to CP160 to achieve the CP concentration of CP180. The ADG and gain:feed ratio (G:F) from day 7 to 21 were highest at CP180. Reduced neAA concentrations in CP160 decreased ADG and G:F. Supplementation of Asp+Asn, Glu, and Glu+Gln to CP160 increased ADG and G:F, but not to the level found for CP180. Compared with CP160, addition of Asp increased G:F but not ADG. Supplementation of Asp+Asn caused higher ADG and G:F than supplementation of Asp alone. The N-utilization efficiency was highest at CP160 and at CP160 supplemented with Ala, Pro, and Glu. Lower N-utilization efficiency was found at CP180 than at CP160, without and with supplemented neAA. The treatment containing NH4Cl presented the lowest ADG, G:F, and N-utilization efficiency. These results showed that individual supplementation of Asp+Asn, Glu, and Glu+Gln partly compensates for the growth-reducing effects of very low CP diets. Supplementation of NH4Cl as NPN source is not suitable for broiler chickens.

Two broiler lines, Line A and Line B, were fed experimental diets from 22 to 42 d with objectives to determine effects of digestible amino acids (AA) to metabolizable energy ratios on feed intake (FI), performance, and processing yield. Experimental diets were formulated to 3,150 kcal/kg with 5 levels of digestible lysine (dLys)-80, 90, 100, 110, and 120% of recommended AA level giving g dLys/Mcal values of 2.53, 2.85, 3.17, 3.48, and 3.80, respectively. All other AA were formulated to a fixed ratio to dLys. A total of 4,050 chicks were utilized in each trial (9 replicate pens for each AA level and each line; 45 chicks/pen) conducted twice: one in hot environmental temperature (HT) (24 h mean ∼85.3 °F; 80.9% RH) and another in cool environmental temperature (CT) (24 h mean ∼71.6 °F; 61.7% RH). Results showed that FI was not impacted by dietary AA levels in HT for both lines. Higher FI (P < 0.05) was observed in CT for lower dietary AA levels (<100% AA level) for both lines, with overall higher FI occurring in Line B. Higher FI for Line B was also accompanied by higher body weight in HT and CT. Treatment diets had quadratic effects on average daily gain (ADG), feed conversion ratio (FCR), and processing yields (breasts and tenders) in both HT and CT, with broilers in CT performing better (P < 0.05). The optimal response values for ADG in HT and CT were 89.72 g and 113.44 g occurring at 120 and 109.5% AA level, respectively. The optimal response values for FCR in HT and CT were 1.79 and 1.58 occurring at 120 and 117.5% AA level, respectively. The optimal response values for breast meat yield in HT and CT were 575.9 g and 776.5 g occurring at 112.6 and 114.5% AA level, respectively. The optimal response values for tender meat yield in HT and CT were 119.8 g and 154.9 g occurring at 120 and 115% AA level, respectively. Line A had a higher breast and tender yield % (of live weight) for both environmental temperatures which correlated to body composition data with higher % protein mass and % digestible AA retention. In this study, findings indicated that effects of increased digestible AA density on FI, performance, and processing yield are specific to strain and grow-out temperature, but the optimum response was attained for both lines with diets containing 110 to 120% AA levels (3.48-3.80 g dLys/Mcal) during the 22 to 42 d finisher period.

Crude protein (CP) levels in commercial broiler (Gallus gallus) diets, optimized for maximum yield production vs. feed cost, have only begun to be assessed for impact on immune function. In order to study immune effects of dietary CP levels, different starter phase (day 1-14) diets were fed to 230 Ross 708 male broiler chicks randomly assigned at 1 day of age into two treatment groups. Group 1: Standard diet (STD) contained 3,000 kcal AMEn/kg energy and 23.78% CP; and Group 2: Reduced crude protein diet (RCP) contained 3,000 kcal AMEn/kg energy and 21.23% CP. From day 15-35 a common standard grower/finisher diet (3,150 kcal AMEn/kg energy and 22.18% CP) was allocated to both groups. Zymosan, a glycan derived from yeast cell walls that binds to TLR 2 and Dectin-1, was used for intra-abdominal challenge. Results demonstrated that a reduced crude protein starter diet (21.23 vs. 23.78% CP) between age 1-14, while maintaining the same levels of metabolizable energy and essential amino acids, did not affect broilers growth performance or lymphoid organ weights (P > 0.05). Interestingly, basal leukocyte levels in the RCP group significantly (P < 0.01) increased in the blood compartment at d35 in the unchallenged birds. Significant enhancements to leukocyte infiltration into the abdominal cavity were also detected post-immune challenge with zymosan (day 14 and day 35; P < 0.01). Post-challenge levels of TNF-α, IL-1ß, and CXCL8 gene expression cells collected from the abdominal cavity were not affected by the diets (P > 0.05). Moreover, dietary treatments did not influence percentage of ROS producing cells in the abdominal cavity (P > 0.05). To our best knowledge, this is the first study that reports the impacts of reduced crude protein diet on the innate immune response of poultry to an acute inflammation model in the abdominal cavity. Overall, our results highlight that reduced crude protein diets can be used without negatively impacting broiler performance and may enhance the capacity of broilers to recruit leukocytes upon infection.

Responses of broiler chickens to dietary glycine equivalent (Glyequi) are affected by dietary cysteine and choline. Hence, this study investigated interactive effects among dietary Glyequi, cysteine, and choline on the growth of broiler chickens. Male Ross 308 broiler chickens were maintained in 105 metabolism units (10 birds/unit) from days 7 to 22. Excreta were collected in 12-h intervals from days 18 to 21. Blood was sampled on day 22 (1 bird/unit). Five levels each of Glyequi (9-21 g/kg), cysteine (2-5 g/kg), and choline (0.5-1.7 g/kg) were tested under 15 diets in 7 replicates each following a fractional central composite design. Another diet was provided to five metabolism units (15 birds/unit) to measure prececal amino acid digestibility. Data were evaluated using neural networks. The gain:feed ratio (G:F) increased with digestible Glyequi intake. Differences between low and high digestible cysteine intake were low. Effects of choline intake on G:F were low. Nitrogen-utilization efficiency (NUE) was high (≥77%), with low variation among treatments. Plasma metabolites varied among treatments and indicated that metabolism of Glyequi, cysteine, and choline was influenced. These findings showed that interactive effects of dietary Glyequi, cysteine, and choline on growth were small, possibly because NUE was barely influenced.

The objective of this study was to compare the standardized ileal digestibility (SID) of amino acids (AA) in 3 poultry by-products including hydrolyzed feather meal (HFM), flash dried poultry protein (FDPP), and poultry meal (PM) and also a meat and bone meal (MBM) between broiler chickens and pigs. Experimental diets consisted of 4 diets containing each test ingredient as a sole source of nitrogen and a nitrogen-free diet. In experiment 1, 416 male broiler chickens with a mean initial body weight (BW) of 705 ± 100 g were allotted to 5 diets with 8 replicate cages per diet in a randomized complete block design with BW as a blocking factor at day 18 posthatching. After 5 d of feeding experimental diets, birds were euthanized by CO2 asphyxiation, and ileal digesta samples were collected from distal two-thirds of the ileum. In experiment 2, 10 barrows with a mean initial BW of 22.1 ± 1.59 kg were surgically fitted with T-cannulas at the distal ileum and allotted to a duplicate 5 × 4 incomplete Latin Square design with 5 diets and 4 periods. Each period lasted for 7 d including 5 d of adaptation and 2 d of ileal digesta collection. Data from experiments 1 and 2 were pooled together and analyzed as a 2 × 4 factorial arrangement with the effects of species (broiler chickens or pigs) and 4 experimental diets (HFM, FDPP, PM, or MBM). There were interactions (P < 0.05) between experimental diets and species in the SID of His, Lys, Thr, Trp, Val, and all dispensable AA except Tyr. In broiler chickens, the SID of Lys in FDPP (73.3%) was greater (P < 0.05) than in HFM (55.7%) but was lower (P < 0.05) than in MBM (86.5%), which was not different from PM (78.7%). In pigs, however, the SID of Lys in FDPP and PM (70.0 and 70.1%, respectively) were greater (P < 0.05) than in HFM (39.0%) but were lower (P < 0.05) than in MBM (79.2%). Broiler chickens fed FDPP and PM had lower (P < 0.05) SID of His, Thr, and Trp than those fed MBM; however, there was no difference in the SID of His, Thr, or Trp among pigs fed FDPP, PM, or MBM. The SID of Val in MBM was greater (P < 0.05) than in the other test ingredients for broiler chickens, but there was no difference in the SID of Val among test ingredients for pigs. Pigs had greater (P < 0.05) SID of Ile and Met than broiler chickens. In conclusion, the pattern of differences in the SID of His, Lys, Thr, Trp, and Val, but not the other indispensable AA, among poultry by-products and MBM were different between broiler chickens and pigs.

Research has shown that methionine+ cysteine (M+C) requirements may be higher when chickens are infected with Eimeria app. In a 4 × 2 factorial design, broilers (11 to 21 D) were fed one of 4 corn-soybean meal-based diets containing either 0.6, 0.8, 0.9, or 1.0% standardized ileal digestible (SID) M+C; on day 14, broilers from each diet were gavaged with either phosphate-buffered saline (PBS) or a commercial coccidiosis vaccine (at 100 × vaccine dose) which provide a mixture of live Eimeria acervulina, Eimeria maxima, and Eimeria tenella oocysts. Growth performance was recorded from day 11 to 21. Plasma and intestinal luminal samples were collected on days 14 and 21. Intestine lesion scores and fecal oocyst counts were conducted on day 21. Regardless of dietary SID M+C levels, compared to PBS gavaged broilers, the Eimeria-challenged broilers had (1) decreased (P < 0.05) body weight gain (BWG), feed intake (FI), and gain-to-feed ratio (G:F); (2) increased (P < 0.05) intestinal lesion scores and fecal oocyst counts; (3) increased (P < 0.05) plasma anti-Eimeria IgG, and intestinal luminal total IgA and anti-Eimeria IgA concentrations; and (4) increased (P < 0.05) levels of duodenum luminal gamma interferon (IFN-γ) and interleukin-10 (IL-10), as well as jejunum and cecum luminal IFN-γ concentrations. Regardless of Eimeria challenge, when compared to 0.6% SID M+C, broilers fed ≥0.8% SID M+C had (1) increased (P < 0.05) BWG, FI, and G:F and (2) increased (P < 0.05) levels of jejunum luminal total IgA. After Eimeria challenge, broilers fed 0.8% SID M+C had increased (P < 0.05) levels of jejunum luminal anti-Eimeria IgA compared to broilers fed diets containing 0.6 and 1.0% SID M+C. Collectively, in 11- to 21-D broilers, the growth suppression caused by Eimeria infection could not be mitigated by further increasing dietary M+C alone ≥0.8%. Further research should investigate interactions between dietary M+C and other nutrients for support of immune function and growth in pathogen-challenged broilers.

A total of 294 male, off-sex Ross 308 chickens were offered 7 dietary treatments with crude protein (CP) contents of 210, 195, 180, and 165 g/kg. One of the four 165 g/kg diet was consistent with the higher protein diets and 3 were modified to investigate the effects of increased methionine levels, pre-pellet inclusion of whole maize, and whey protein concentrate in reduced-CP broiler diets. There were 7 replicate cages, 6 birds per cage, from 14 to 35 D post-hatch. The average feed conversion ratio (FCR) of birds offered 210, 195, 180 g/kg CP diets was 1.555 which was superior (P < 0.05) to the 1.608 FCR of their 165 g/kg counterparts. The transition from 210 to 165 g/kg (diet 4) CP diets linearly increased (P < 0.001) relative fat-pad weights from 8.64 to 14.62 g/kg. The same transition linearly increased jejunal and ileal starch digestibility coefficients (P < 0.001), metabolizable to gross energy ratios (ME:GE) ratios (P < 0.001) and nitrogen (N)-corrected apparent metabolizable energy (AMEn) (P = 0.001) but did not influence N retention. Starch:protein disappearance rate ratios increased linearly (P < 0.001) from 2.68 to 3.82 in the jejunum and from 1.76 to 2.94 in the ileum following dietary CP reductions. Ileal disappearance rate ratios were quadratically related to FCR (r = 0.486; P < 0.005) and linearly related to relative fat-pad weights (r = 0.663; P < 0.001) where both parameters were disadvantaged by widening ratios. The transition from 210 to 165 g/kg crude protein diets linearly increased the average digestibility coefficient of 17 amino acids from 0.459 to 0.594 in jejunum and from 0.744 to 0.790 in the ileum. The present study demonstrates that dietary CP can be reduced from 210 to 180 g/kg without negatively influencing broiler performance but the further reduction to 165 g/kg compromised FCR. However, the three modifications to the 165 g/kg CP diet failed to enhance broiler performance.

The crude protein (CP) content of 4 iso-energetic, maize-based diets containing 11.00 g/kg digestible lysine was reduced in gradations from 200 to 156 g/kg with increasing inclusions of synthetic, or unbound, essential amino acids. A constant dietary electrolyte balance (DEB) of 230 mEq/kg was maintained, but a second 156 g/kg CP diet had a DEB of 120 mEq/kg, and energy densities of the 156 g/kg CP diet were reduced in the sixth and seventh treatments. Each of the 7 dietary treatments were offered to 7 replicate cages (6 birds/cage) or a total of 294 Ross 308 off-sex male broilers from 14 to 35 D posthatch. Reductions in CP from 200 to 156 g/kg did not influence weight gain but quadratically increased feed conversion ratio (FCR) and linearly increased relative abdominal fat-pad weights and feed intakes. The reduction in DEB did not influence growth performance but did adversely influence some amino acid digestibilities. Reducing energy density by 100 kcal/kg did not influence growth performance of birds offered the 156 g/kg CP diet but numerically reduced fat-pad weights. The transition from 200 to 156 g/kg CP diets generally enhanced jejunal and ileal amino acid digestibility coefficients but had diverse effects on free amino acid concentrations in systemic plasma with a remarkable 116% increase in threonine. Starch:protein disappearance rate ratios linearly increased in the jejunum and the ileum following the same transition, and these expanding ratios were related to heavier fat-pads and compromised FCR. This study indicates that reductions in dietary CP from 200 to 172 g/kg supported by inclusions of unbound essential amino acids do not compromise growth performance, but a further reduction to 156 g/kg CP significantly increased FCR. Both heavier relative fat-pad weights and inferior FCR were related to expanding starch:protein disappearance rate ratios, which suggests condensed dietary starch:protein ratios may advantage birds offered reduced CP diets.

The aim of this study was to indentify the involvement of leptin receptors (LepR) in astrocytes in hippocampal synaptic transmission and plasticity and metabolism. To this end we used a genetic mouse model (GFAP-LepR-/-) of specific LepR ablation in GFAP positive cells and recorded excitatory postsynaptic potentials (fEPSPs) within the CA1 area. Glutamate (Glu) uptake and the expression of Glu transporters (EEAT3, GLT-1 and GLAST) and enzymes involved in Glu metabolism (glutamine synthase, GABA decarboxylase 65 and 67) were quantified. Modifications in the expression of GFAP, the glucose transporter (GLUT)-1, and the monocarboxylate transporters MCT-2 and MCT-4, were also analyzed. The results show that depletion of LepR in GFAP positive cells reduced basal synaptic transmission within the CA1 area and impaired N-methyl-d-aspartate (NMDA)-evoked long-term depression (NMDA-LTD). Hippocampal slices from GFAP-LepR-/- mice displayed lower Glu uptake efficacy together with up-regulation of GLT-1, glutamine synthase, GFAP and GLUT-1. In conclusion, astrocyte LepRs are involved in the maintenance of Glu homeostasis and Glu neurotransmission within the hippocampus. Our findings support a role of hippocampal LepRs in synaptic plasticity, which could have an impact on memory and learning processes.

However, while the negative impact on the liver of these diets has been an issue of interest, systematic research on the effect of HFDs are lacking. Objective: To characterize the overall impact of HFDs on both molecular and morphological signs of liver remodeling. Methods: A study was conducted on male C57BL/6J mice to assess the effect of 4- and 8-week HFDs (60% kcal from fat) on (I) liver steatosis and fibrosis, and (II) expression of factors involved in inflammation and angiogenesis. Results: After an 8-week HFD, vascular endothelial growth factor type-2 receptor (VEGF-R2) and fatty acid translocase/trombospondin-1 receptor (CD36) were overexpressed in liver tissue of mice given HFDs. These changes suggest impaired liver angiogenesis and occurred together with (I) increased GPR78-BiP and EIF2alpha phosphorylation, suggesting endoplasmic reticulum stress, (II) induction of Col1a1 gene expression, a marker of fibrosis, and (III) increased CD31 immunolabeling, consistent with active angiogenesis and fibrosis. Conclusion: Our data show that very HFDs promote a rapid inflammatory response, as well as deregulation of angiogenesis, both consistent with development of liver fibrosis


Antecedentes: El hígado graso no alcohólico (HGNA) es una enfermedad hepática que ocasiona fibrosis y se genera por la ingesta de dietas ricas en grasa. Aunque los efectos nocivos de este tipo de dietas son de gran interés, no son muy abundantes los estudios sistemáticos sobre las consecuencias que su consumo puede tener en el hígado. Objetivo: Evaluar los efectos de una dieta rica en grasa sobre el remodelado hepático, tanto a nivel morfológico como molecular. Métodos: Se utilizaron ratones macho C57BL/6J tratados durante 4/8 semanas con una dieta que contenía un 60% de las kilocalorías procedentes de grasa sobre: 1) la aparición de esteatosis y/o fibrosis hepática y 2) la expresión de factores implicados en procesos de inflamación y angiogénesis. Resultados: Tras 8 semanas de dieta se observó un incremento en el receptor del factor de crecimiento vascular endotelial tipo 2 (R2-FCVE) y en la translocasa de ácidos grasos (CD36). Estos cambios, que sugieren que los procesos angiogénicos del hígado están alterados, fueron simultáneos a: 1) un aumento del GPR78-BiP y de la fosforilación de EIF2alpha, marcadores de estrés del retículo endoplásmico, 2) la inducción en la expresión del gen de colágeno Col1a1, indicador de fibrosis y 3) un incremento de células inmunofluorescentes para CD31 compatible con procesos de angiogénesis y de fibrosis. Conclusiones: Nuestros resultados muestran que las dietas con alto contenido en grasa inducen la rápida activación de respuestas inflamatorias, así como alteraciones en la angiogénesis, siendo ambos procesos compatibles con el desarrollo de fibrosis hepática

BACKGROUND: The minimum to which dietary crude protein (CP) level for broiler chickens can be reduced without decreasing growth and the glycine equivalent (Glyequi) concentration required are not known. The plasma metabolome might reflect dietary influences on physiological processes. OBJECTIVE: The aim of this study was to investigate the effect of 3 low CP levels with 4 Glyequi concentrations on growth and characteristics of nitrogen excretion, and to identify plasma metabolome variations. METHODS: Male Ross308 broiler chickens were provided 1 of 12 dietary treatments in 84 metabolism cages (10/cage) from days 7 to 21. Three diets with 163 (CP163), 147 (CP147), and 132 (CP132) g CP/kg were formulated, each containing 12, 15, 18, and 21 g Glyequi/kg. Essential amino acid concentrations were the same in all diets. Animals and feed were weighed on days 7 and 21 to determine average daily gain (ADG) and gain:feed ratio (G:F). Excreta were collected from days 18 to 21 to analyze nitrogenous components, and blood was obtained on day 21 to conduct a metabolome analysis. RESULTS: Two-factor ANOVA showed significant interaction effects for ADG, G:F, and nitrogen efficiency (P < 0.001). Reduction of CP decreased ADG and G:F, and increased nitrogen efficiency. Glyequi supplementation increased ADG (by 7.9 g/d) and G:F (by 0.07 g/g) at CP132. The ADG (by 2.4 g/d) at CP147 and G:F (by 0.02 g/g) at CP147 and CP163 increased up to 15 g Glyequi/kg. Multivariate statistical analysis showed an influence of Glyequi on plasma acylcarnitine and lysophosphatidylcholine concentrations, and a decrease of plasma phosphatidylcholine and sphingomyelin concentrations with reduced CP. CONCLUSIONS: These results suggest that a nutrient other than Glyequi limited growth when CP was reduced from CP163 to CP147, and that the response of broiler chickens to Glyequi is dependent on the dietary CP level. Plasma metabolites indicate dietary influences on the physiological state of the animals.

A Box-Behnken designed study was completed to predict growth performance, carcass characteristics and plasma hormone and metabolite levels as influenced by dietary energy, amino acid densities and starch to lipid ratios in male broiler chickens. The design comprised three dietary energy densities (11.25, 12.375 and 13.5 MJ/kg), three digestible lysine concentrations (9.2, 10.65 and 12.1 g/kg) and three starch to lipid ratios (4.5, 12.25 and 20.0) in broiler diets based on maize and soybean meal. Each of thirteen dietary treatments was offered to 10 replicates of 15 birds per replicate floor pen or a total of 1,950 Ross 308 male broiler chickens from 21 to 35 days post-hatch. Increasing dietary energy decreased feed intake with a quadratic relationship between feed intake and dietary standardised ileal digestible (SID) Lys concentrations, where increasing SID Lys initially increased and then depressed feed intake. Increasing dietary amino acid density increased body weight gain and carcass weight; however, dietary energy did not influence body weight gain, carcass and breast meat weight. Feed efficiency was positively influenced by energy and amino acid densities but negatively influenced by starch to lipid ratios and energy and amino acids had more pronounced impacts than starch to lipid ratios. This study indicated that both energy and amino acid densities regulate feed intakes in broiler chickens. Body weight gain of modern broiler chickens is more responsive to amino acid densities; nevertheless, dietary energy density continues to play an important role in protein utilisation, as reflected in significantly reduced plasma uric acid levels.

The risk assessment in terrestrial environments has been scarcely studied for mixtures of organic contaminants. To estimate toxicity due to these compounds, an ecotoxicological test may be done with the appropriate organism and biomarker. Photosynthesis is principally performed at photosystem II, and its efficiency is affected by any environmental stress. Consequently, the measure of this efficiency may be a good indicator of toxicity if different parameters are employed, e.g., the quantum efficiency of photosystem II and the photochemical quenching coefficient. We did a series of assays to determine the toxicity of two organic contaminants, ibuprofen and perfluorooctanoic acid, using a higher plant (Sorghum bicolor). The results showed more toxicity for the perfluorinated compound and greater sensibility for the quantum efficiency of photosystem II. Regarding the binary combination, three methods were applied to calculate EC50: combination index, concentration addition, and independent action. Synergistic behavior is the principal toxicological profile for this mix. Therefore, the combination index, which considers interactions among chemicals, gave the best estimation to determine risk indices. We conclude that the inhibition of photosynthesis efficiency can be a useful tool to determine the toxicity of the mixtures of organic pollutants and to estimate ecological risks in terrestrial environments.
